چکیده

Sertoli cells under the influence of Follicle Stimulating Hormone are known to provide a microenvironment for the growth and differentiation of germ cells. Follicle Stimulating Hormone regulates proliferation of Sertoli cells only during the early neonatal period and during adult period it regulates functional parameters like Transferrin, Androgen binding protein and other factors needed for germ cell differentiation and how this is achieved is not known. In an attempt to understand the differential action of Follicle Stimulating Hormone on the Sertoli cells, we employed the approach of neutralization of endogenous Follicle Stimulating Hormone in neonatal rats for a very limited period and study the effect on the regulation of gene expression by Differential Display RT-PCR analysis. One of the transcripts was found to be down regulated following neutralization of endogenous Follicle Stimulating Hormone in the neonatal rats and it shared identity with Intracisternal A particle element, an endogenous retrovirus. Often integration of Intracisternal A particle into host genes and its regulation result in growth factor independence suggesting a role in proliferation. Interestingly, the expression of Intracisternal A particle transcript is not regulated by Follicle Stimulating Hormone in the adult rat Sertoli cells suggesting a possible reason for stimulation of proliferation of Sertoli cells by Follicle Stimulating Hormone only in

Involvement of peroxisome proliferator-activated receptors in the estradiol production of ovine Sertoli cells

Peroxisome proliferator-activated receptors (PPARs) are nuclear receptors of transcription factors composed of three family members: PPARα, PPARβ/δ and PPARγ. This study was aimed to evaluate the role of PPARs in the estradiol production via follicle stimulating hormone (FSH) in the ovine Sertoli cells. At the first step, transcripts of PPARα, PPARβ /δ and PPARγ were evaluated by quantitative r...

Follicle-stimulating hormone amplifies insulin-like growth factor I-mediated activation of AKT/protein kinase B signaling in immature rat Sertoli cells.

FSH and IGF-I are both important determinants of testicular development and Sertoli cell function. The present studies were performed to determine the actions of FSH and IGF-I on PI3K/AKT protein kinase signaling in immature rat Sertoli cells. Primary cultures of rat Sertoli cells were prepared from 10-d-old rats. After 7 d in culture, Sertoli cells were treated with IGF-I, FSH, or IGF-I plus F...

Role of winged helix transcription factor (WIN) in the regulation of Sertoli cell differentiated functions: WIN acts as an early event gene for follicle-stimulating hormone.

Members of the winged helix transcription factor family are known to regulate epithelial cell differentiation by regulating cell-specific gene expression. rWIN is a newly discovered member of the winged helix family shown to be present in the adult rat testis. In the testis the human homolog of rWIN, HFH-11, was localized to the germ cells (i.e. spermatocytes and spermatids) undergoing spermato...

O-13: Na+/K+-ATPase Alpha1 Isoform Mediates Ouabain-Induced Expression of Cyclin D1 and Proliferation of Rat Sertoli Cells

Background: Novel roles for the interaction of cardiotonic steroids to Na+/K+-ATPase have been established in recent years. The aim of the present study was to investigate the intracellular signaling events downstream the action of ouabain on Na+/K+-ATPase in Sertoli cell obtained from immature rats. Treatment of Sertoli cells with ouabain (1 μM) induced a rapid and transient increase in the ex...

Sodium chlorate induces undersulfation of cellular proteoglycans and increases in FSH-stimulated estradiol production in immature rat Sertoli cells.

The functional influence of cell proteoglycan (PG) undersulfation on estradiol synthesis by immature rat Sertoli cell cultures was investigated by using sodium chlorate, an inhibitor of the active sulfate donor for sulfotransferases. The addition of sodium chlorate to 20-day-old rat Sertoli cell cultures abolished [35S]-sulfate incorporation into neosynthesized PG and consequently reduced the r...
